Nodal segments of coffee cv. Caturra Amarelo established in vitro were pre-cultured in MS medium without the growth regulators or with BAP at 6 mg/litre. After 180 days, the nodal segments were inoculated in MS medium supplemented with BAP at 0, 6, 9, 12 and 15 mg/litre, in the first experiment. In the second experiment, the nodal segments were inoculated in MS medium containing TDZ at 0, 0.05, 0.15, 0.45 and 1.35 mg/litre. The pH of the medium was adjusted to 5.8 and solidified with 0.7Summary: agar. Explants were then transferred to the growth room at 27.1°C temperature, 16 h photoperiod and 32 AM m-2 s-1 light intensity for 120 days. There was a residual effect of BAP added to the pre-culture medium. Addition of BAP (9-10 mg/litre) and TDZ (0.15-1.35 mg/litre) to the sub-culture medium increased the total number of sprouts, number of sprouts between 0.5 and 1.0 cm, number of sprouts larger than 1.0 cm, and sprout fresh matter weight.
